Fluorometric Determination of Cyanide with o-Phthalaldehyde and Taurine

Abstract

Several fluorogenic reagents such as p-benzoquinone and pyridoxal have been developed for the determination of cyanide ion. However, their applicability is often limited by the low sensitivity and by the necessity of the complicated procedure. In our previous study on the determination of sulfide ion, it was found that cyanide ion showed a fluorescence in the presence of taurine and o-phthalaldehyde (OPA). This fluorescence seems to be due to the formation of a fluorescent isoindole derivative (Fig. 1) because cyanide is known to give 1-cyano-2-methylisoindole when treated with methylamine and OPA. The present work aimed to establish a simple and sensitive fluorometric method for determining cyanide ion by using the OPA reaction.
